CHAPTER TITLE : Applications of RNAi in C . elegans research
نویسندگان
چکیده
The discovery of RNA interference in C. elegans in 1998 has established this nematode as a key model to study RNAi mechanisms. Meanwhile, RNAi has been extensively applied to study a variety of basic biological questions in worms. In C. elegans, the dsRNA required to trigger RNAi can be administered by injection, by soaking, and importantly, by feeding. The delivery of dsRNA through food and the existence of two RNAi feeding libraries allow for large scale RNAi approaches. Thus, RNAi libraries have been used to screen for genes related with processes as diverse as embryonic development, longevity, fat accumulation, DNA damage response, axon guidance or synapse structure, among others. Moreover, since RNAi facilitates the inactivation of more than one gene at the time in the same animal, RNAi assays are being applied to uncover genetics interactions among genes and therefore there is a significant and ongoing progress in identifying components of genetics pathways and networks. Here we review successful applications of RNAi as tool in C. elegans research, and discuss trends in using such tool to shed light into several biological research fields.

RNA Interference in Practice
In the short time since its elucidation, RNA interference (RNAi) has quickly emerged as the premier tool in which to study gene function. RNAi provides a simple and predictable yet extremely powerful and specific method in which to knock down gene expression in virtually any cell type or whole organism. The major advance in RNAi technology, particularly for mammalian systems, came with the disc...
متن کاملE. coli OxyS non-coding RNA does not trigger RNAi in C. elegans
The discovery of RNA interference (RNAi) in C. elegans has had a major impact on scientific research, led to the rapid development of RNAi tools and has inspired RNA-based therapeutics. Astonishingly, nematodes, planaria and many insects take up double-stranded RNA (dsRNA) from their environment to elicit RNAi; the biological function of this mechanism is unclear. Recently, the E. coli OxyS non...
متن کاملUsing RNAi in C. elegans to Demonstrate Gene Knockdown Phenotypes in the Undergraduate Biology Lab Setting
RNA interference (RNAi) is a powerful technology used to knock down genes in basic research and medicine. In 2006 RNAi technology using Caenorhabditis elegans (C. elegans) was awarded the Nobel Prize in medicine and thus students graduating in the biological sciences should have experience with this technology. However, students struggle conceptually with the molecular biology behind the RNAi t...
متن کاملRNA Interference in Caenorhabditis elegans.
RNAi has become an essential tool in C. elegans research. This unit describes procedures for RNAi in C. elegans by microinjecting with dsRNA, feeding with bacteria expressing dsRNA, and soaking in dsRNA solution, as well as high-throughput methods for RNAi-based screens.

The recently completed Caenorhabditis elegans genome sequence allows application of high-throughput (HT) approaches for phenotypic analyses using RNA interference (RNAi). As large phenotypic data sets become available, "phenoclustering" strategies can be used to begin understanding the complex molecular networks involved in development and other biological processes. The current HT-RNAi resourc...
